There is a rgb ssd, a strip on the bottom, 2 rgb fans and 2 rgb ram.

However you want to do it. Keep in mind that the vent positions are there for a reason (unless it's a shitty case). Proper airflow should be a priority. This does not mean, "open hole = more flow"

Would it not be better to mount standoff screws onto the case frame and put the holes in the acrylic?

This makes the most sense to me.

It’s sheet metal, so if any cut is not done with a properly designed blade, or sharp enough it’s gonna come out in an irreversibly bent and burred condition. It would make it quite hard to replace the “hole” with an acrylic panel.
