It is literally where the word Lesbian comes from. It meant the people of Lesbos way before it were associated with homosexual women.
But some people from Lesbos are more lesbian than others.

Probably not. "Sappho" has Greek origins, "sapphire" is Hebrew is origin. They're likely not connected.
